Master Thesis "Characterization and immobilization of diamine oxidase"

 

IMAGINE

 

  • The consumption of food containing biogenic amines is reported to be responsible for many pharmacological effects (diarrhea, headache, asthma, hypotension) which lead to several types of foodborne disease, including histamine poisoning, mainly from ingestion of certain species of fish (scombroid poisoning), and tyramine toxicity (cheese reaction).

 

ENGAGE

 

  • The master student will determine the response behaviour of DAO from various animal and plant sources towards biogenic amines using different methods and bioassays. Enzyme activity will be determined as well as protein content. It is expected that enzymes from different origin have different capability to oxidize and thus degrade biogenic amines, such as putrescine, histamine, spermine, cadaverine and others. In addition, the stability of the enzyme will be studied at different temperatures. Protectants might be added to improve stability. Alternatively, DAO will be encapsulated in alginate beads using the Büchi Encapsulator.
  • After a solid literature review the master student will determine the response behaviour and stability of free and encapsulated diamine oxidases from various sources and finally deliver a set of fully characterized enzymes to be implemented in the multiplex test for detection of biogenic amines.

 

ACHIEVE

 

  • We will develop a multiplex diagnostic tool that is capable of reliably and simultaneously measuring biogenic amines with the overall goal to provide a validated detection system for further investigations.
  • This tool is based on diamine oxidase (DAO) which catalysis the degradation of biogenic amines under formation of hydrogen peroxide and ammonia.

What to expect:
